The thrombospondins are a family of extracellular matrix molecules
that mediate the cell-to-cell and cell-to-matrix interactions that
are involved in development, wound healing and neoplasia.
In Drosophila, the single thrombospondin gene mediates the attachment
of muscles to tendon. These effects are mediated through interactions
with cell surface receptors, such as integrins, proteoglycans,
calreticulin, CD36 and CD47. Thrombospondin-1 (TSP-1) is unique among
the members of the thrombospondin gene family in its ability to bind
and activate latent transforming growth factor-beta. TSP-1 also
regulates proliferative, adhesive, migratory and invasive properties
of various cell types. In the tumor microenvironment, TSP-1 promotes
the migration of tumor cells and cancer-associated fibroblasts but
inhibits the migration of endothelial cells. These activities have
prompted the development of TSP-1 mimetics for the inhibition of
tumor growth.
