siLentMer[TM] Validated Dicer-substrate siRNA duplexes
The siLentMer siRNAs are a new class of long dsRNAs (27-nt) with
altered end structure capable of initiating an enhanced, more specific
silencing. They are functionally tested by RT-qPCR to select only
those that produce a reduction of >85% in target mRNA expression using
5 nM of siRNA. Gene silencing is sustained at 80-85% knockdown for 6-9

These two studies provide insight into the role of CtBP1/BARS in
macropinocytosis, a dynamin-independent endocytic pathway. While Urs
Greber and colleagues show that CtBP1-dependent macropinocytosis is an
entry route for human adenovirus type 3, Alberto Luini and coworkers
investigated the function of CtBP1 in growth factor-stimulated
macropinocytosis.

The fission yeast homologue of CENP-B, Abp1, regulates directionality
of mating-type switching
Directed recombination at the mating-type locus requires
heterochromatin-mediated long-range interactions. Here, Azorín and
colleagues identify the first factor specifically required for
spreading of the recombination-promoting complex along
heterochromatin, while being dispensable for heterochromatin formation
itself.
