BONE MORPHOGENETIC PROTEIN ANTAGONISTS: MIGHTY MORPHIN SMALL MOLECULES

A screen for chemical antagonists of the bone morphogenetic protein
(BMP) pathway has identified dorsomorphin as a selective inhibitor
of BMP receptor-mediated SMAD1/5/8 phosphorylation.

Rock2
http://ealerts.nature.com/cgi-bin24/DM/y/eiUj0Xztnp0H2W0BkiZ0EC

Rock2 is a serine/threonine kinase that is activated by association
with RhoGTP. Activation of Rock2 leads to the stabilization of
filamentous actin and increased myosin ATPase activity. These events
promote the formation of contractile actin-myosin bundles (often
called stress fibers) and integrin-containing focal adhesions.
Through modulating actin-myosin contractility, Rock2 has a
significant role in the regulation of cell morphology, motility and
adhesion.

THERAPEUTICS: SMAC IT TO THEM!
Compounds that activate or mimic the pro-apoptotic protein SMAC
(second mitochondrial-derived activator of caspase) cause regression
of human lung cancer xenografts by increasing apoptotic signaling
through tumor necrosis factor receptor 1 (TNFR1).

DEVELOPMENTAL BIOLOGY: RETINOIC ACID PASSES THE MORPHOGEN TEST
The anterior-to-posterior retinoic acid (RA) concentration gradient
is established by the opposing effects of a locally acting,
RA-induced degradation enzyme and fibroblast growth factor (Fgf),
which represses RA degradation.

G-PROTEIN-COUPLED RECEPTORS: CRYSTAL CLEAR
A collection of papers describing the crystal structure of the
G-protein-coupled beta2-adrenergic receptor (beta2AR) may facilitate
the development of drugs that specifically interfere with
ligand-receptor binding of related GPCRs.

DRUNKEN FLIES GET HYPERSEXUAL
http://ealerts.nature.com/cgi-bin24/DM/y/eiUj0Xztnp0H2W0Bkka0EL

From the annals of insect biology comes a cautionary tale for those
recovering from their post-New Year's celebration: heavy boozing has
been shown to send male fruitflies into a lusty fog. The findings
could be used to further establish a fly model system for the study
of alcoholism.
