We develop stem cell technology for potential use in diabetes research and treatment. Our research has led to a patent-pending method for generation of adult stem cells from cells isolated from tissues. While embryonic stem cells are capable of differentiation into all of the various cells of the body, adult stem cells are more limited in differentiation capacity generally including the cell types that comprise the tissue of origin. Adult stem cells are now used for transplantation therapy of blood disorders, e.g., leukemia. Our present focus is on the application of adult stem cells to therapeutic replacement of endocrine organs that produce hormones essential to physiological regulation of the body. While use of embryonic stem cells involves controversial ethical issues and complex differentiation procedures, we use tissue-derived adult stem cell lines with demonstrated differentiation capacity for our research. In addition to treatment of diabetes, our novel stem cell generation technology may provide therapeutic options to other diseases or conditions resulting from death or degeneration of specific cells including, for example, age-related hearing loss, macular degeneration, muscular dystrophy, heart attack, and congestive heart failure, Parkinson’s disease, etc.

The Company has generated 29 adult human pancreatic stem cell lines that are distinct from the limited number of embryonic stem cell lines that are presently approved for federally funded research. Some of these lines have been extensively expanded in cell culture and have demonstrated self-renewal properties by growth through 100 to 200 population doublings (PD) while mortal cells typically do not expand beyond 50 population doublings. Also, we have developed methods of differentiation that result in expression of biomarkers characteristic of alpha and beta cells suggesting that the cell lines have at least multipotent differentiation capacity. Some markers of non-pancreatic cell lineage are expressed in these cells raising the possibility of differentiation into other cell types. We manufacture select human cell lines and related products according to GMP standards for use in stem cell and drug discovery research. Our research products include two human cell lines of pancreatic origin and supporting products including optimized growth and cryopreservation media. These lines are Mesenchymal, attachment-dependent cells that exhibit properties of human Mesenchymal stem cells, affording researchers several unique opportunities.

Toxicity Screening Tools

AV-101

VistaGen's lead drug candidate, AV-101, is a potential first-in-class "GlyB site" antagonist of the NMDA-receptor, entering Phase 1 clinical development for epilepsy and neuropathic pain markets. AV-101 is expected to address the limitations of previous NMDA-receptor antagonists by providing high efficacy and expected low adverse side effects. AV-101 will potentially prevent or delay the onset and reduce the severity of epileptic seizures. Concurrently, AV-101 minimizes neural toxicity that accompanies the pathological activation of NMDA-receptors, which are associated with seizures, inflammation and brain damage. The TriStem Group has been set up by Mr.Ghazi Dhoot and Dr. Ilham Saleh Abuljadayel to further research and develop its retrodifferentiation technology which is used to create stem cells from mature adult cells. The retrodifferentiation technology is based on the results of 12 years of research and currently TriStem is the only company that owns, and is in the process of commercialising, this technology. The main advantage of TriStem's new process over existing methods of harvesting stem cells is that it provides large quantities of stem cells which are by definition DNA matched and, therefore, bypasses the political and ethical issues concerned with therapeutic cloning.

TriStem believes that there is a wide ranging potential for clinical, pharmaceutical and biotechnological applications for its proprietary stem cell technology. For example, novel compounds could be screened for their retrodifferentiation ability in order to manipulate a specific cell type in the body and such cells can be studied for research purposes to better understand stem cell biology.

Stem cells are being investigated as therapeutics for a wide variety of applications. TriStem will initially evaluate the clinical potential of its retrodifferentiation stem cell technology in aplastic anaemia, thalassemia and leukaemia patients that would ordinarily require a blood marrow transplant (BMT). Leukaemia, a disease which resulted in an estimated 22,100 deaths in the United States alone in 1999, is also a disease area with a wide unmet clinical need and a large potential for expedited clinical development. TriStem will also in due course explore the application of its technology in other areas of oncology such as lymphoma, myeloma and solid tumours. In addition, the technology should over time be employed in the treatment of a much wider range of illnesses such as spinal cord injury, diabetes and heart disease.

ThromboGenics Ltd is a privately held company founded in Dublin, Ireland in 1998, focused on discovery and development of biotherapeutics for treatment of vascular diseases (including cardiovascular, cerebrovascular and peripheral vascular disease). ThromboGenics' founder and CEO is Désiré Collen, M.D., Ph.D., a world-renowned expert in cardiovascular disease. Prof. Collen directs the Molecular and Cardiovascular Medicine Group at University of Leuven, Belgium, which provides much of the initial research for ThromboGenics programs. Professor Collen's laboratory was the first to produce clinical supply of tissue plasminogen activator (tPA), one of the most effective drugs for thrombolytic therapy of acute myocardial infarction (heart attack). This proprietary molecule is licensed to Genentech in return for royalties on worldwide sales.

Recently, our development of improved ES cell derivation methods, have generated us the possibility to derive ES cells from any mouse strain. These improved ES cell derivation methods even allow the derivation of ES cells from mice strains that were previously considered as non-permissive for ES cell derivation. All derived cell lines have proven germ-line transmission capability at high passage numbers. Among the future research aims of Thromb-X, are the derivation of embryonic stem cells from non-murine species (rabbit, rat, pig), the nuclear transfer technique to produce genetically modified rabbits, and the establishment of pronuclear injection in pigs.
